Design, Synthesis and Anti-Tubercular Activity of Isoniazid Derivatives

The aim of this study was to develop novel isoniazid derivatives, which can be utilized as antitubercular agents. The compounds were synthesized by reaction between the hydrazine unit of isoniazid with an appropriate lipophilic moiety. The compounds obtained were characterized on the basis of their melting points, R-f value, Fourier Transform Infra Red (FTIR) Spectra, H-1-Nuclear Magnetic Resonance (H-1-NMR) Spectra, C-13- Nuclear Magnetic Resonance (C-13-NMR) Spectra, Elemetal Analysis, and the Mass spectra. The novel compounds were assessed for their anti-tubercular activity by reported method against four strains of the Mycobacterium. The minimum inhibitory concentration of the active compounds was also determined. The anti-tubercular activity data revealed that most of the compounds have higher MIC values as compared to the standard drug isoniazid. Two compounds, namely 3b and 3c, exhibited very good anti-tubercular activity against all four strains of the Mycobacterium, which was comparable to the standard drug isoniazid. It has been concluded that the incorporation of a lipophilic chain at the position-2 of the pyridazine ring along with a halogen group, preferably a fluorine atom, at the p-position of the phenyl ring in this series of compounds increase the anti-tubercular activity of the compounds. However, this assumption cannot be generalized because in the present study the anti-tubercular activity was assessed against four strains of Mycobacterium. Accordingly, further studies are recommended to assess the anti-tubercular activity of these two compounds, 3b and 3c, against other strains of the Mycobacterium.

Electron stimulated desorption from condensed pyrimidine and pyridazine

Low energy electron (LEE) interactions and the formation of transient negative ions play a dominant role in radiation-induced dissociation of condensed-phase biomolecules (e.g. in radiotherapy). Here we present data on the LEE-induced dissociation and desorption of the DNA/RNA-base and radiosensitizing agent analogues pyrimidine and pyridazine. Vapors of each molecule were condensed on either a Pt or Ar substrate to form a multilayer film or a submonolayer molecular target, respectively. These were irradiated with electrons of 0-80 eV and the desorbing anionic and cationic fragments analysed via time of flight mass spectrometry. The detected cations are the same species seen in gas-phase mass spectra, albeit of differing relative intensity. Anion yield functions exhibit strong maxima, indicating that transient negative ions contribute significantly, via dissociative electron attachment (DEA), to molecular dissociation below 20 eV. For both molecules, the < 5 eV shape resonances, seen experimentally and predicted by theory, do not result in fragment desorption. The main anionic fragments are H- and CN- for both molecules, additionally the fragments C-, CH- C2H- and CHN- desorb from pyrimidine and C- and C2H- from pyridazine, with some resonances lying above the ionization limit. Pyrimidine shows higher anion desorption yields than pyridazine for all species except H-. The anion signal also comprises dipolar dissociation (DD), investigated in both anionic and cationic yield functions. From analysis of anion and cation yields, fragmentation pathways are suggested. The direct ionization pathway provides information on the appearance energies for cations and their production processes in condensed phase. In an endeavor to develop antitumor agents, we made a credible survey regarding synthesis, structure, and pharmacological assay of novel pyridazine derivatives, so that 2-((6-(4-chloro-3-methylphenyl)pyridazin-3-yl)oxy)acetohydrazide 3 was utilized as scaffold to build novel compounds 4-19 by reaction with various electrophilic reagents, followed by determination and explanation atropisomerism phenomena and tauomerism ratio such as keto-enol and lactam-lactim tautomers for some synthesized compounds. In vitro, these compounds were screened for antitumor efficacy versus two cell lines, namely, hepatocellular carcinoma and mammary gland breast cancer, by using MTT assay. Among the examined compounds, compound 16 was exhibited promising potent activity (IC50 = 8.67 +/- 0.7 mu M) versus HepG2 cell line. Meanwhile, compounds 3 and 16 were manifested the very highest efficacy (IC50 = 5.68 +/- 0.6 and 9.41 +/- 0.9 mu M) versus MCF-7 cell line.

Identification of imidazo[1,2-b] pyridazine TYK2 pseudokinase ligands as potent and selective allosteric inhibitors of TYK2 signalling

As a member of the Janus (JAK) family of non-receptor tyrosine kinases, TYK2 mediates the signaling of pro-inflammatory cytokines including IL-12, IL-23 and type 1 interferon (IFN), and therefore represents an attractive potential target for treating the various immuno-inflammatory diseases in which these cytokines have been shown to play a role. Following up on our previous report that ligands to the pseudokinase domain (JH2) of TYK2 suppress cytokine-mediated receptor activation of the catalytic (JH1) domain, the imidazo[1,2-b] pyridazine (IZP) 7 was identified as a promising hit compound. Through iterative modification of each of the substituents of the IZP scaffold, the cellular potency was improved while maintaining selectivity over the JH1 domain. These studies led to the discovery of the JH2-selective TYK2 inhibitor 29, which provided encouraging systemic exposures after oral dosing in mice. Phosphodiesterase 4 (PDE4) was identified as an off-target and potential liability of the IZP ligands, and selectivity for TYK2 JH2 over this enzyme was obtained by elaborating along selectivity vectors determined from analyses of X-ray co-crystal structures of representative ligands of the IZP class bound to both proteins.

Discovery of novel triazolo[4,3-b]pyridazin-3-yl-quinoline derivatives as PIM inhibitors

PIM kinase family (PIM-1, PIM-2 and PIM-3) is an appealing target for the discovery and development of selective inhibitors, useful in various disease conditions in which these proteins are highly expressed, such as cancer. The significant effort put, in the recent years, towards the development of small molecules exhibiting inhibitory activity against this protein family has ended up with several molecules entering clinical trials. As part of our ongoing exploration for potential drug candidates that exhibit affinity towards this protein family, we have generated a novel chemical series of triazolo[4,3-b]pyridazine based tricycles by applying a scaffold hopping strategy over our previously reported potent pan-PIM inhibitor ETP-47453 (compound 2). The structure-activity relationship studies presented herein demonstrate a rather selective PIM-1 /PIM-3 biochemical profile for this novel series of tricycles, although pan-PIM and PIM-1 inhibitors have also been identified. Selected examples show significant inhibition of the phosphorylation of BAD protein in a cell-based assay. Moreover, optimized and highly selective compounds, such as 42, did not show significant hERG inhibition at 20 mu M concentration, and proved its antiproliferative activity and utility in combination with particular antitumoral agents in several tumor cell lines. Luminescent thermometer based on Eu3+/Tb3+-organic-functionalized mesoporous silica

In this work we investigate a mesoporous silica (MS) decorated with dipyridyl-pyridazine (dppz) ligands and further grafted with a mixture of Eu3+/Tb3+ ions (28.45%:71.55%), which was investigated as a potential thermometer in the 10-360K temperature range. The MS material was prepared employing a hetero Diels-Alder reaction: 3,6-di(2-pyridyl)-1,2,4,5-tetrazine was reacted with the double bonds of vinyl-silica (vSilica) followed by an oxidation procedure. We explore using the dppz-vSilica material to obtain visible emitting luminescent materials and for obtaining a luminescent thermometer when grafted with Eu3+/Tb3+ ions. For the dppz-vSilica@Eu,Tb material absolute sensitivityS(a) of 0.011K(-1) (210K) and relative sensitivityS(r) of 1.32 %K-1 (260K) were calculated showing good sensing capability of the material. Upon temperature change from 10K to 360K the emission color of the material changed gradually from yellow to red.

Synthesis and structural studies of 3,6-disubstituted-bis-1,2,4-triazolo-[4,3-b][3 ‘,4 ‘-f]pyridazines

Synthesis of a series of 3,6-disubstituted-bis-1,2,4-triazolo-[4,3-b][3′,4′-f]pyridazines (4) was accomplished by the oxidative intramolecular cyclization of 6-arylidenehydrazino-3-aryl-1,2,4-triazolo[4,3-b]pyridazine (3) using iodobenzene diacetate (IBD), as a green oxidant, in dichloromethane at room temperature. The compounds 3 and 4 were characterized by IR, NMR (H-1 and C-13), mass spectral data and elemental analyses. X-ray crystal analysis of sterically strained 3,6-di-(2′-fluorophenyl)-bis-1,2,4-triazolo-[4,3-b][3′,4′-f]pyridazine 4f and 3,6-di-(4′-fluorophenyl)-bis-1,2,4-triazolo-[4,3-b][3′,4’-f]pyridazine 4g indicated that pyridazine ring has twisted conformation leading to nonplanar tricyclic core. Both compounds crystallized in orthorhombic P2(1)2(1)2(1) space group, containing one single molecule per asymmetric unit. The studies reveal that the compound 4f is associated with weak, centrosymmetric F center dot center dot center dot F interactions (distance of 2.882(3) angstrom), with cis geometry, between adjacent molecules which are responsible for the formation of chains along a axis. Additionally, compounds 4 were screened for their cytotoxic activity against the human cervical carcinoma (HeLa) cell line using MTT assay, however, with not much significant activity.
